The drugs taken from eucalyptus leaves have been used long since in medicine as anti-inflammatory, bactericidal and regenerative agents. The leaves of eucalyptus contain chlorophyll and its precursors which have a good regenerative activity. Besides these components, the eucalyptus volatile oil holds cineol as a major ingredient which have a bactericidal activity. As results, remedies are made on this basis have the ability to accelerate the healing of open wounds. These properties make such remedies suitable for the treatment of very widespread burns. The drugs are manufactured from this raw material can be used for treatment of following diseases: burn, sluggish wounds, venous stasis ulcers.
The plants of Eucalyptus genus have a limited of growth area which is located largely in Australia and New Zeeland. At the same time practically, all known herbs can be sources of chlorophyll e.g. such widespread plant as a nettle. Moreover, in addition to Eucalyptus sp. the cineol contains in a sage (Salvia officinalis) which is a prevalent plant of family Lamiaceae. It would appear reasonable that remedies taken from aforesaid plants will possess by similar or nearly action.
This work was aimed research of chemical composition of plants and pharmacological activity of phytoremedies taken from chosen herbs. The leaves of Eucalyptus viminalisLabill., Salvia officinalis L. and Urticadioica L. which were collected in Krasnadar and Siberia regions of Russia have been chosen as objects of this investigation.
Collected leaves were dried up and crushed. The extracts were made by using the extraction from raw material by ethanol. Extracts were made by way of macerating during 24 hours on temperature 25 ̊C and in the proportion a raw material: solvent — 1 :50. Derived extracts were filtered after completing of extract process and have been used in the future for analysis of their chemical composition. Qualitative analysis of composition was made by using TLC method with comparing deriving chromatogram with certified reference standards of above-mentioned compounds. As a result, it was established that three major compounds prevail in chemical composition of researched raw material -chlorophyll, cineol and chlorogenic acid.
The content of these components of chemical composition of the total extract was determined by the spectrophotometry method.
The made analysis of spectra extremums showed extract from eucalyptus and sage leaves have the chemical composition with such major components as chlorophyll and cineol.Ethanol extract from nettle leaves in the wavelength rangefrom 200 to 400 nm have sharp distinctions from other extracts. This fact can be explained by existence of chlorogenic acid in composition of this extract.
That is the reason of appearing in total spectra of second maximum on
326 nm and of drift of first maximum from 276 nm to 292 nm. At the same time the extremums which are characteristic for cineol weren`t discovered to UV spectra of nettle extract.
The content of chlorophyll, cineol and chlorogenic acid was calculated according to the absorption of UV-light with character wavelength in comparing with solutions of reference standards (tabl.1).
Table 1 Content of bioactive compounds in investigated raw material
Plant | Amount, % | ||
chlorophyll | cineol | chlorogenic acid | |
Eucalyptus | 3,2 | 0,65 | — |
Sage | 2,8 | 0,12 | 0,03 |
Nettle | 4,4 | — | 2,35 |
The ointments have been created on the basis of the manufactured extracts with usage of lanolin as a ground. The application of lanolin possessing an amphiphile properties allowed to make a homogeneous mixture including hydrophilic (chlorogenic acid) and lipophilic (chlorophyll) compounds.
The research of pharmacological activity was conducted in rats with the model of thermal burn [1]. As a result, it has been established that healing with the usage of the ointments received from Eu.viminalis and S.officinalis accelerates twice in comparison with control. As for ointment on the basis of U.dioica that significant acceleration of process of healing hasn’t been revealed.
To conclude, the chlorophyll and cineol complex in comparison with the chlorophyll and chlorogenic acid complex has bigger regenerative activity.
It is possible to assume that antiseptic properties of cineol are favorable for healing open wounds.
